Kirilenko started playing tennis at age of just five. She won 2002 US Open Junior Girls Tournament in singles category. She began her professional career in 2001 but started playing in WTA events in 2002. In 2004, she reached to the first final of her career in a tournament at Hyedrabad where she lost to Australian player Nicole Pratt by 6-7,1-6. She also won her first doubles title at DFS Classic (Birmingham) the same year where she partenered with Maria Sharapova.
Kirilenko won her first singles title at China Open in 2005 beating Anna-Lena Gronefeld in the final by a score of 6-3,6-4. Another double title came in her way at AIG Japan Open Tennis Championships partenering Gisela Dulko. At the end of 2005, she ranked 25 in singles WTA Ranking and 27 in doubles Ranking. Year 2006 was not very good for her as she dropped to No. 30 in singles ranking and 35 in doubles ranking at the end of the year. She also not won any title in this year. 2007 and 2008 proved very fruitful for Kirilenko as she won four singles titles and three doubles titles during this period. Her biggest win in doubles was at Cincinati Masters titles in 2008 where she partenered Nadia Petrova and beaten the pair of Hsieh Su-wei and Yaroslava Shvedova in the final by 6-3,4-6,10-8. She reached her best Ranking of No. 18 in 2008. She won three more doubles titles in 2009 and 2010 including her second win at 2010 Cincinati Masters. This time her partener was Victoria Azarenka. The pair beats Lisa Raymond and Rennae Stubbs in a tough final by 7-6,7-6.

Kirilenko improved her game in doubles during this period. She ranked No. 5 in women's doubles ranking which is also her best ranking in doubles event. She also won two Premier Mandatory Tournaments - first at 2011 Madrid Masters and second at 2012 Miami Masters. In 2011 Australian Open she reached the quarter-finals of the singles event which is also her best performance at any Grand Slam Event. She also achieved her best performance in doubles event of any Grand Slam in that tournament finishing as the runner-up of the tournament. She lost to the pair of Gisela Dulko and Flavia Pennetta.
